Suppression of Mismatch Amplification

		MutS binds to mismatch forming a very stable template:protein complex.
		Aac DNA polymerase is unable to extend beyond the complex. These templates are consequently eliminated from the amplification pool. Only full-match templates amplify.

Stringent hybridization of primers provides one check-point to promote faithful amplification. However in all DNA amplification processes, mismatch priming occurs with some frequency. Only in SmartAmp can amplification of the mismatched products be fully suppressed (by MutS).
